Zusammenfassung: | The invention belongs to the technical field of gene engineering and particularly relates to primers and a method for unknown RNA mycovirus genome cloning. The primers comprise a forward primer and a complementary primer with nucleotide sequences shown in SEQ ID No.1 and SEQ ID No.2 respectively. The method is used for unknown RNA mycovirus genome cloning by the primers to obtain dsRNA whole-length cDNA sequences of unknown viruses. The forward primer is designed according to iresine herbstii virus sequences, has no similarity to known fungus and plant virus sequences and avoids nonspecific amplification during amplification of virus sequences; meanwhile, joint sequences are shorter than known joint sequences and have high binding efficiency; a random primer is added besides the complementary primer in reverse transcription in the virus genome amplification process, more complete reverse transcription of virus genomes can be guaranteed, and the primers are adaptable to amplification of RNA genomes of mycoviru |
