The invention relates to a gel microsphere modified anti-fouling oil-water separation ultrafiltration membrane and a preparation method thereof. The gel microsphere is prepared by using vinyl monomersrespectively containing a carboxyl group and an amide group on the side groups, firstly a random copolymer is formed by free radical polymerization, a physically cross-linked network structure is formed through hydrogen bonding supra-molecular self-assembly between the molecular chain side groups, and the particle size of the microsphere is adjustable within the range of 50-500 nm; and the surface of a base film is subjected to the carboxylation (-COOH) modification by an ultraviolet light initiated graft polymerization method, and is connected with the amide groups (-CONH2) on the surface ofthe microsphere by the hydrogen bonding action.
