Zusammenfassung:The invention relates to a B-type dye-decolorizing peroxidase and a mutant and application thereof, in particular to a B-type dye-decolorizing peroxidase gene dyp derived from Pseudomonas sp.Q18. A recombinant engineering bacterium is constructed for the dyp gene, expression induction, separation and purification are executed to obtain the B-type dye-decolorizing peroxidase PmDyP. According to theinvention, on the basis of an amino acid sequence of the PmDyP, amino acid residues at the positions 125, 142 and 188 are mutated to obtain the PmDyP-5c mutant with improved catalytic efficiency. Themutant has the good degradation effect on lignin raw materials, and has application and popularization values in the utilization process of biomass resources. 本发明涉及一种B型染料-脱色过氧化物酶及其突变体和应用。具体涉及一种来源于假单胞菌Pseudomonas sp.Q18的B型染料-脱色过氧化物酶基因dyp,对dyp基因构建重组工程菌,并诱导表达、分离纯化获得B型染料-脱色过氧化物酶PmDyP。本发明在PmDyP氨基酸序列的基础上,对第125、142和188号位的氨基酸残基进行突变,获得催化效率提高的PmDyP-5c突变体。本发明的突变体对木质素原料具良好的降解效果,在生物质资源的利用过程具有应用和推广价值。
