The method comprises the steps of chromatographic condition and system adaptability testing, preparation of a reference solution, preparation of a test solution, determination, and the like. The chromatographic conditions and the preparation method of a test product are optimized; two chiral compounds of the amygdalin can be separated fundamentally by adopting a UPLC method; and after the D-amygdalin and L-amygdalin are separated, the content of the D-amygdalin is determined, so that the accuracy of content determination is greatly enhanced. 本发明公开了种桃仁配方颗粒中D-苦杏仁苷的含量测定方法，包括色谱条件与系统适应性试验、对照品溶液的制备、供试品溶液制备、测定法等步骤。本发明通过优化色谱条件及供试品的制备方法，采用UPLC法能从根本分离苦杏仁苷两种手性化合物，将D-苦杏仁苷与L-苦杏仁苷分离后再测定D-苦杏仁苷的含量，大大增强了含量测定的准确性。
