Zusammenfassung:The present invention discloses a method for extracting and separating nerve cells in a mouse brain. After the isolated mouse brain is disinfected with alcohol, the isolated mouse brain is placed in apre-cooled sterile balanced salt solution D-hanks; the brain is chopped with a scalpel, the chopped brain is digested with a 0.15% (m/v) papain solution, the D-hanks buffered saline solution containing deoxyribonuclease I is used to blow cells, and the cells pass through a 100 [mu]m filter screen; then cell separation fluid percoll differential centrifugation is used to remove myelin sheath proteins to obtain a single cell suspension; a red blood cell lysate is used to remove red blood cells; and various types of the nerve cells are obtained by purification by using magnetic beads of miltenyi. 本发明公开了种小鼠脑中神经细胞的提取分离方法,将离体小鼠脑部用酒精消毒后置于预冷无菌平衡盐溶液D-hanks中;用手术刀将脑切碎,用0.15%(m/v)的木瓜蛋白酶液消化后,用含脱氧核糖核酸酶I的D-hanks缓冲盐溶液吹打细胞后,过100μm滤网;然后用细胞分离液percoll差速离心去除髓鞘蛋白,得到单细胞悬液;用红细胞裂解液去除其中血红细胞;利用美天旎miltenyi的磁珠beads纯化得到各种类型的神经细胞。
