The method includes the steps of: acquiring the medical image of a to-be-operated part; reconstructing a virtual three-dimensional model according to medical image, also marking different body tissues in the virtual three-dimensional model, and setting a positioning guide platefixed on one of the body tissues in the virtual three-dimensional model; and making an entity model of the positioning guide plate in the virtual three-dimensional model so as to realize augmented reality positioning of the to-be-operated part by actual spatial position matching of the positioning guide plate's entity model and a virtual model in the virtual three-dimensional model. The method and device provided by the invention can conveniently and effectively realize augmented reality positioning of the to-be-operated part, assist the operation to avoid injuring nerves, blood vessels and other important parts, and
