The method comprises the following steps: diluting the molasses alcohol waste mash till a COD value is 1800 to 2000 mg/L, adjusting the pH value to be 6.8 to 7.0 with a NaOH solution with the concentration being 1 mol/L, and performing sterilization for 20 min at 121 DEG C, so asto obtain a culture medium; inoculating microalgae into the culture medium, performing light shake-flask culture, ensuring that the light intensity is 3800 to 4200 lux, the shaker speed is 145 to 155r/min, and the culture temperature is 23 to 27 DEG C, and measuring total nitrogen, total phosphorus and COD values in the culture medium.
