Zusammenfassung: | The invention relates to a gene mutation detection technology, and concretely relates to a method for improving the detection sensitivity of gene mutation. The method comprises the following steps: S1, PCR amplification of a targeted DNA sequence; S2, purification of a PCR product; S3, extension of a mutant specific primer; and S4, hybridization and enrichment of mutant gene DNA fragments. The method is different from a conventional ASPE technology used for gene mutation detection, only a mutant forward primer and unmarked dNTPs are added to the extension reaction system of the mutant specificprimer; and the mutant gene DNA fragments are enriched through the denaturation and renaturation processes of nucleic acid after the hybridization of the mutant gene DNA fragments.
