The probe combination comprises probes shown as SEQ ID NO. 1 to SEQ ID NO. 4 which include a 5S rDNA probe, a 5.8S rDNA probe, a 18S rDNA probe and a 25S rDNA probe respectively; the probe combination and the method are used for carrying out in-situ hybridization on plant chromosomes, can be used for capturing chromosomes with a relatively high resolution ratio and fluorescent in-situ hybridization signals, and are suitable for the in-situ hybridization of the chromosomes of various plants; the probe combination is an oligonucleotide probe and has the characteristics of low productioncost, convenience for utilization, simplicity in operation and the like.
