The invention provides a lactic acid bacteria high-density fermentation culture medium, a preparation method and application of the preparation method. The culture medium is prepared from protein enzymatic hydrolysate, yeast powder and part of salt, wherein the enzymatic hydrolysate is prepared by hydrolyzing skimmed milk powder and soy isolate protein by using alcalase protease. The lactic acid bacteria high-density fermentation culture medium has a specific formula including the skimmed milk powder (6 percent), the soy isolate protein (2 percent), glucose (3 percent), barley flour (2 percent), yeast powder (1.8 percent), NaCl (0.23 percent), VC (0.3 percent), MgSO4.7H2O (0.02 percent), MnSO4.H2O (0.005 percent) and sodium citrate (0.02 percent). The culture medium is used for performinghigh-density culture on lactobacillus plantarum, lactobacillus rhamnosus and lactobacillus paracasei; in the stable period of culture, the viable count in a culture solution can be up to 1-1.5 x 10 cfu/mL; the viable count in
