The method includes the steps that 1, agaricus bisporus compost is prepared and includes straw, fowl manure, calcium superphosphate, lime and urea, and the raw materials are mixed; 2, the mixed raw materials are cut into pieces, water is added, and then the cut raw materials are mixed evenly and soaked for 2-3 days; 3, the soaked mixed raw materials are fermented at a ventilating position, and scattered with the lime during fermentation, and stirring is performed one time at a time interval; 4, fermented compost is loaded into a box for seeding, and a layer of thin film is arranged on the compost; 5, the compost and strains are moved into a culture room to be arranged on a rack for hypha culture, and temperature is kept at 25-27.5 DEG C.
