Zusammenfassung: | The invention relates to an in-situ diffraction apparatus of protein crystal and an in-situ diffraction method thereof. The in-situ diffraction apparatus comprises a layer of double adhesive isolationstrip, two layers of high-molecular films, two layers of glass plates, and two layers of transparent adhesive tapes. The double adhesive isolation strip is used for forming a microscale reservoir having a certain thickness, which is used for growth of the protein crystal. The X-ray permeability of the high-molecular film is used for realizing the in-situ diffraction of the crystal. The in-situ diffraction apparatus has fixing effect for the high-molecular film, ensures the direct installation at the various diffraction apparatuses, ensures no requirement of dismounting of a glass plate whilea diffraction experiment is carried out, realizes a diffraction purpose without configuration of specific hardware equipment, and realizes the in-situ operation from crystal growth to diffraction datacollection.
