The method comprises the following steps: firstly, building a two-chamber type microbial fuel cell; adding a graphene oxide solution in an anode chamber of the microbial fuel cell; reducing graphene oxide at an anode by utilizing a reduction function of an anode microorganism; carrying out interaction self-assembly with microorganisms to form a graphene biological anode; carrying out electrode reverse operation on the cell; converting the graphene biological anode into a graphene biological cathode through selection of the microorganisms; building the microbial fuel cell with the double graphene bio-electrodes by using the prepared graphene biological anode and the graphene biological cathode as two electrodes so as to environmentally friendly prepare the microbial fuel cell with the graphene bio-electrodes.
