Zusammenfassung: | The invention discloses a method of utilizing RNA nucleic acid aptamer and nanogold to detect theophylline in serum. The method includes following steps: pretreating biological and clinical samples, hybridizing an embedment probe and target miRNA, connecting a capture sequence onto a surface modified ELISA plate, performing nucleic acid hybridization to form an ELISA plate-embedment probe-miRNA compound, checking through RNase ONE ribonuclease, and using a chemiluminescence method to detect target miRNA content and theophylline content. According to a pretreatment scheme in Direct PCR technology, biological cells or clinical lung cancer samples are chemically and biologically treated to degrade protein and DNA ingredients and to release total RNA therein without RNA extraction, so that experimental process is simplified, detection time is shortened, and precious clinical tumor samples can be saved.
