The device is mainly used for simulating the stratum heterogeneity from the centimeter level by combining an actual stratum in a laboratory. The device is composed of a cuboid-shaped artificial rock core having a size of 1*1*30 cm and having differentpermeability values, a first screwed joint, a second screwed joint, an artificial centimeter-level poured heterogeneous rock core, an inlet valve, an outlet valve and a rock core bracket. The manufacturing method of the device comprises the following steps: obtaining the distribution law of actual measurement data of the actual stratum, determining the permeability distribution of the centimeter-level rock core according to the law, and simulating the permeability distribution law of the stratum in the centimeter-level rock core in order to simulate the stratum heterogeneity.
