The invention relates to a detachable lake on-site test device and an operating method thereof. The device includes an acrylic plate assembly and a connecting assembly. The acrylic plate assembly includes a plurality of square acrylic plates. The connecting assembly includes corner connecting members, edge connecting members and middle connecting members. Adjacent two edges of each corner connecting member, any three edges of each edge connecting member and four edges of each middle connecting member are provided with buckling connection structures having same structures. The acrylic plates are provided with buckling structures corresponding to the position of the buckling connection structures and having structures matched with the buckling connection structures.
