The preparation method comprises the steps of activating human plasminogen (Plg) by virtue of a plasminogen activator (PA) at room temperature so as to generate plasmin, adding a terminating agent to terminate reaction, and carrying out separation and purification, wherein the terminating agent is capable of inhibiting the degradationof plasmin. 本发明涉及种人纤维蛋白溶解酶的制备方法。本发明在常温条件下用纤维蛋白溶解酶原激活剂(Plasminogen Activator，PA)激活人纤维蛋白溶解酶原(Plasminogen,Plg)生成纤维蛋白溶解酶，激活后加入终止剂终止反应，同时该终止剂可抑制纤维蛋白溶解酶降解，然后进行分离纯化即可。
