Zusammenfassung:The invention relates to the field of cell engineering, in particular to a PEG4000 mediated potato protoplast transformation method and application thereof. The method includes: well mixing prepared potato protoplast with PEG4000 solution being 40v/v% in concentration and vector plasmids connected with target genes, treating in dark at the room temperature for 5-7min, removing supernatant, and subjecting precipitates to resuspension culture to obtain a transformant. The method is convenient and simple in operation, low in cost and low in harms to protoplasts, and the transformation rate reaches about 70%. 本发明涉及细胞工程领域,具体而言,涉及种PEG介导的马铃薯原生质体转化方法及其应用。该方法包括:将制备好的马铃薯原生质体与40v/v%浓度的PEG溶液、连接有目的基因的载体质粒混匀,在室温下黑暗处理5~7min后弃去上清并将沉淀重悬培养即得转化子。本方法操作简便,成本相对较低,转化率可达70%左右,且对原生质体伤害较小。
