The invention provides a PCR reaction device. According to the PCR reaction device, a first through hole and a second through hole are formed in a semi-closed channel to realize communication between the PCR reaction device and outside, wherein a reaction system can enter a PCR reaction tube through one of the through holes; the other through hole through which the reaction system does not pass realizes inside-outside gas pressure balance, under a semi-closed state, of the PCR reaction device, so that connection between the PCR reaction device and a reaction device at the front end and conveying of the reaction system are realized. The invention further provides a using method of the PCR reaction device.
