The preparation method provided by the invention comprises the following steps: with 5-methoxy-2-tetralone as a raw material, carrying out hydrogenation reduction so as to obtain 5-methoxy-1,2,3,4-tetrahydro-2-naphthol; and then with p-chlorophenol acetate as an acyl group donor, porcine pancreatic lipase (PPL) as a resolution catalyst and acidic resin D006 as a racemization catalyst, carrying out dynamic kinetic resolution so as to obtain (2R)-5-methoxy-1,2,3,4-tetrahydro-2-naphthol. The method has the characteristics of simple operation, high product yield, good optical purity of the product, etc., and is of great guidance and application value to production of and research on (2R)-5-methoxy-1,2,3,4-tetrahydro-2-naphthol.
