The nucleotide sequence of the thermophilic beta-1, 4-xylanase-His fusion protein gene is shown as SEQ ID NO.1, and the construction method comprises the steps of: step A: cultivation of Geobacillus thermoleovorans; step B: genomic DNA extraction and fusion protein gene sequence construction; step C: primer design and fishing of a thermophilic beta-1, 4-xylanase-His fusion protein target gene by polymerase chain reaction (PCR); step D: construction of a recombinant expression vector; and step E: transformation of the recombinant vector into a host cell.
