Zusammenfassung: | The invention provides a preparation method of 3-cyano-5-methoxy-2-methylindole, comprising the following steps: mixing 5-methoxy-2-methylindole, K4[F3(CN)6].3H2O, a catalyst, a catalyst promoter and an organic solvent to obtain a reaction system, the catalyst being glycine palladium chloride complex, and providing green atom-economic cyanation reaction by catalytic C-H activation to obtain the 3-cyano-5-methoxy-2-methylindole.Compared with the prior art, the preparation method of the invention uses the glycine palladium chloride complex as the catalyst replacing traditional Pd(OAc)2, and the yield of 3-cyano-5-methoxy-2-methyindole can be greatly increased.Results of an embodiment of the invention show that the yield of the 3-cyano-5-methoxy-2-methylindole by the method provided herein is higher than 78%, up to 83.90%.
