Zusammenfassung:The invention discloses a multiplex PCR (polymerase chain reaction) detection method of tobacco Ty-2 gene and Ty-5 gene. The detection method comprises the following steps: 1) extracting to-be-detected tobacco leaf DNA from tobacco leaves; 2) by taking the DNA extracted from the tobacco leaves as a template, conducting multiplex PCR amplification on a to-be-detected sample by an established multiplex PCR reaction system by virtue of primers of Ty-3 and Ty-5; 3) conducting electrophoresis detection on a PCR product; 4) identifying whether the to-be-detected tobacco sample contains the Ty-2 gene and Ty-5 gene or not; and the like. The detection method disclosed by the invention is used for rapidly identifying whether tobacco yellow leaf curl resistant genes Ty-2 gene and Ty-5 exist in a tobacco material or not; therefore, the detection method is beneficial for screening disease-resistant gene materials when pyramiding breeding is conducted on the two genes.
