Zusammenfassung: | The invention belongs to the field of animal gene engineering, and particularly relates to a coded high-hydrolysis-activity PLE (pig liver carboxylesterase) subtype gene. A novel subtype gene with the hydrolysis activity on a general substrate of p-NPA (p-nitrophenyl acetate) being higher than that of the reported PLE subtype gene is obtained through the separation from the liver tissues of health large white pigs; the nucleotide sequence of the gene is shown as SEQ ID NO:1; the coded PLE protein has high hydrolysis activity; function verification proves that the enzyme activity of the PLE protein gene is 13.29U/mg and is much higher than the enzyme activity of seven kinds of reported subtypes. The single-ingredient PLE isozyme with the high catalytic hydrolysis activity is obtained through cloning and expression; the production cost can be greatly reduced; the huge practical value is realized on the organic chemical synthesis industry and the pharmacy. |
