The method comprises the steps of parent hybridization, embryo rescue, backcross, self-cross and identification of Oryza sativa L.ssp.indica and Oryza sativaL.ssp.japonica, wherein the parent hybridization is that Yuanjiang Oryza rufipogon Griff. is used as a male parent, Oryza sativa L.ssp.indica is used as the female parent, and a pollen mixture obtained by mixing anthers of dead Oryza sativa L.ssp.indica with pollens of Oryza rufipogon Griff. according to a mass ratio of 1: 1 is used for pollination; an embryo culture medium comprises modified MS, 8 g/L of agar powder and 10 g/L of sucrose and has the pH of 5.8-6.0. According to the invention, the method greatly improves the success rate of hybridization, rescues the immature embryos obtained through hybridization, and improves the success rate of distant hybridization of cultivated rice and Oryza rufipogon Griff. from 0-3% to 60%-70%; through the synergy of all steps, Oryza sativa L.ssp.indica and Oryza sativaL.ssp.japonica are obtained simultaneously; the method is simple and practicable; a new method and technical support are provided for the cultivation of endemic new strains of Oryza sativa L.ssp.indica and Oryza sativaL.ssp.japonica. |
