ITS2 sequence; conducting PCR amplification by taking a daktulosphaira vitifoliae genome DNA as a template; detecting the amplification result through agarose gel electrophoresis, wherein if the size of the amplification result is the 114bp nucleotide sequence, the sample contains the daktulosphaira vitifoliae; further, conducting real-time fluorescence quantitative PCR analysis on the sample containing the daktulosphaira vitifoliae, and visually conducting qualification and quantification on the content of the daktulosphaira vitifoliae according to the relation graph of the sample hole cycle number and the fluorescence intensity. The PCR technology is utilized for identification of the daktulosphaira vitifoliae at the root parts of the grape nursery stocks for the first time, the difficulty of nursery stock quarantine inspection is solved, the real-time fluorescence quantitative PCR technology is beneficial to further qualification and quantification of the sample by a quarantine department, the propagation and explosion risk of the daktulosphaira vitifoliae can be mastered and judged, and the method has the advantages of being high in sensitivity and accuracy, simple and convenient to operate and small in risk.
