The method includes the steps: firstly measuring light utility efficiency G of a tree crown, crown amplitude D, root breath occupied proportion B, leaf shading rate f and instantaneous photosynthetic rate Pi; calculating annual net carbon Wa of per leaf area; substituting custody coefficient H, total leaf area LAT =S*LAI, S= Pi*D2/4 and per leaf area averagely receiving light intensity value Y=1/(f*LAI) into a formula I, namely CL=H*G*(1-B)*Wa*LAT*Y; calculating a carbon sequestration value of the single plant tree. Based on a gas exchange method, a carbon sequestration calculating model of the single urban plant tree is built by combining apparent characteristics of the tree crown, the light utility efficiency and a custody mode, urban green land carbon sequestration capacity is accurately measured and calculated, and the method has the advantages that the method is high in pertinence, simple to operate, high in accuracy, nondestructive, suitable for multiple urban heterogeneous environments and the like.
