According to the technical scheme, the method comprises the following steps: 1) purifying a PCR segment or a synthetic nucleic acid segment, diluting and determining the concentration; 2) transferring to a 96-hole Golden gate reaction prefabricated slab, and carrying out Golden gate reaction together with the 96-hole Golden gate reaction prefabricated slab; (3) transferring a reaction product to a competent state cell plate, and transforming the Golden gate reaction product; (4) transferring the cell to a large LB solid culture medium, and separating a monoclonal antibody; (5) choosing the monoclonal antibody, culturing, transferring into the 96-hole Golden gate reaction prefabricated slab, and cloning and identifying a Golden gate reaction product; (6) transferring to a deep 96-hole plate, cultivating, extracting cloning plasmids, and determining the concentration of the cloning plasmids; and (7) sequencing the plasmids.
