The method comprises the following steps: performing sterile operation and recording primary urine as VB1, recording middle-section urine as VB2, EPS, and recording primary urine as VB3 after massage; treating an EPS specimen; treating a calculus specimen; culturing the specimen; performing indirect immunofluorescence staining and fluorescence microscope observation; performing transmission electron microscope scanning; performing ultrathin section observation; performing scanning electron microscope observation; performing Dahl McGec-Russel Alizarin red staining, thus obtaining the result determination standard.
