Zusammenfassung: | The invention relates to a method for inducing conformation deformation and unfolding of protein by virtue of a static electric field. According to the invention, common proteins including trypsin, chymotrypsin, glucose oxidase and bovine serum albumin are embedded by phosphate buffer of poly(2-acrylamido-2-methyl-1-propanesulfonate, methylene bisacrylamide and phosphate buffer solution of ammonium persulfate. The structure of the electroactive polymer contains lots of sulfonic acid groups, and under alkaline condition, the sulfonic acid groups carry negative charges, so that an electrogenerated response environment at the periphery of protein molecules is created. The protein is arranged in a fluorescence detection system, fluorescent light with wavelength being 332nm is emitted due to excitation of ultraviolet light with wavelength being 280nm. Once unfolding occurs on the protein, Trp embedded in the structure of the protein is gradually exposed in a solvent, so that intensity of the emitted ultraviolet light changes. An instrument adopted by the method disclosed by the invention is simple and convenient to operate, and the method is expected to be further applied in research on unfolding, refolding and misfolding of the protein. |
