Zusammenfassung: | The invention discloses a method for screening HSF (heat shock transcription factor) target DNA by combining formaldehyde crosslinking/chromatin immunoprecipitation and molecular cloning. According to the method, by combining chromatin immunoprecipitation (ChIP) and molecular cloning technology, arabidopsis thaliana heat shock transcription factor HSF combined target DNA is separated and identified in vivo. The method comprises: firstly combining HSF of heat-shock induced arabidopsis thaliana leaf with target DNA, then performing formaldehyde processing to enable HSF and target DNA to be subjected to covalent crosslinking, then using HSF antibody to performing chromatin immunoprecipitation, performing adaptor-mediated PCR amplification on chromatin immunoprecipitation DNA, then performing molecular cloning, and finally sequencing, analyzing the sequencing result in a genome database, and identifying the position of target DNA fragment in genome and corresponding HSF target gene. The combination of formaldehyde crosslinking/chromatin immunoprecipitation and molecular cloning is an ideal method for screening target DNA of heat shock transcription factor HSF. |
