Zusammenfassung: | The invention discloses a method for inducing hairy roots of scutellaria baicalensis through low-energy ion injection mediated Ri plasmid transformation. According to the method, a low-energy ion injection mediation technique is utilized to efficiently and directionally introduce Ri plasmid into explant of scutellaria baicalensis, the Ri plasmid is prompted to be recombined and integrated with genomes of scutellaria baicalensis, then the purpose of inducing the hairy roots of scutellaria baicalensis is achieved, the hairy roots of scutellaria baicalensis are over grown in form, rich in branch and free of geotropism and can grow rapidly without hormone, baicalin can be stably synthesized biologically, the induction rate can be 20-55%, and the content of baicalin can be 16.47-19.56%. The method disclosed by the invention is not only simple and convenient to operate, but also is wide in application range, and makes the basis for establishing hairy root systems of monocotyledon and gymnospermae. |
