Zusammenfassung: | A SCAR marker of a sweet sorghum high sugar gene adopts sweet sorghum LTR102 as female parent and sorghum 654 as male parent. 400 random primers are screened by applying the BSA (Bull Serum Albumin) method and the RAPD (Random Amplified Polymorphic Dna) technology, wherein the primer S336 is amplified out difference bands; through the proof of coseparation analysis results, three plants S3361119 in 120 offspring plants are recombined, and the recombination rate is 3.3 percent. RAPD segments amplified from the primer S336 are reclaimed to be subjected purifying, clone and testing in sequence; and the target segment is 1119 bp in length through results. The marker is named as S3361119. One pair of specific primers are designed according to the sequence test result; the specific amplification is performed to successfully convert the RAPD marker to the SCAR marker; and the SCAR marker is used for performing the quick PCR (Polymerase Chain Reaction) detection on the F5-generation and part of sorghum and sweet sorg |
