The invention discloses a mouse simulation system on a touch screen, which comprises a mouse simulator, a defining module, an identification module, a control module, a system menu and a simulation cursor. The invention also discloses a mouse simulation method on the touch screen, which comprises the following steps of: a step 1 of starting the mouse simulator; a step 2 of defining positions and operating modes of a left key touch point, a right key touch point, a roller touch point and a multidirectional moving touch area; a step 3 of completing simulating the clicking operation of a mouse and the rolling and clicking operation of a mouse roller and identifying the operation corresponding to a touch behavior; a step 4 of controlling the simulation cursor to execute the operation; and a step 5 of shutting down the mouse simulator.
