The invention provides a method for detecting the transgenic pawpaw strain 16-0-1, comprising the following steps of: providing a pawpaw nucleic acid sample and a primer pair; forming a polymerase chain mixture from the pawpaw nucleic acid sample and the primer pair and carrying out an amplification reaction to obtain an amplification reaction product; and detecting the amplification reaction product, wherein if an amplification segment in a preset size exists, the sample contains the DNA (Deoxyribonucleic Acid) of the transgenic pawpaw strain 16-0-1. The invention also provides a primer for detecting the transgenic pawpaw strain 16-0-1, a probe and a kit. The sequence, the method, the primer and the kit are favorable to meeting the regulatory requirements and intellectual property protection and rapidly sieving homozygous transgenic offspring.
