In the method, a precursor of purine or pyrimidine nucleotide and phosphate ion are used as substrates, glucose is used as a ribose group donor and energy donor, a permeable microbial cell is used, and a chemical micro molecular effect matter is used to control a cofactor regeneration system, a matter energy coupling regeneration system and the enzymatic activity of a metabolic pathway key enzyme, so that the high-efficiency preparation of the nucleotide is realized.
