The invention discloses a method for determining content of total cell wall substances in tobacco leaves, which adopts microwave technology, and selects benzene-ethanol mixture solution as extracting solvent and a refit soxhlet extractor as the extraction device. The method comprises the following steps of: making pulverized sample of tobacco leaves into a package; disposing the package in the refit soxhlet extractor containing benzene-ethanol mixture solution; placing in a refit microwave oven for extracting and heating treatment; adding distilled water for heating treatment; vacuum-filtering the solvent containing the total cell wall substances and drying to obtain the total cell wall substances in the tobacco leaves; and calculating the content.
