Zusammenfassung: | 1354525 Inducing diuresis : calcitonin compositions ARMOUR PHARMACEUTICAL CO 19 April 1971 [17 April 1970] 26563/71 Heading A5B Diuresis of sodium acid water without excessive potassium depletion is obtained by administering to non-human animals an effective quantity (e.g. 0.1 to 50 micrograms/Kg body weight of natural or synthetic fish ultimobronchial calcitorin having, in the substantially pure state, a potency of 3500 MRC units. Compositions which may be used may be in conventional forms suitable for oral, parenteral, topical, intravaginal and rectal administration and may contain conventional diluents, carriers and excipients. A method of obtaining suitable calcitonin (which in substantially pure form should have a potency of 3500 MRC units per mg) comprises homogenizing ultimobronchial tissue with 0.1N HCl, centrifuging, subjecting the extract to ultracentrifugation, lyophilising the supernatant, taking up in formic acid, adding trichloracetic acid, allowing to stand removing the precipitate, treating with an ion exchange resin in acetate gauge form to remove acid, diluting and lyophilising. Further purification may be by fractionation on "Sephadex" (Trade Mark) and C.M. "Sephadex" columns. |
