SCREENING OF CONFORMATION SPECIFIC BINDING PARTNERS OF GPCRS

A method of producing a conformational specific binding partner of a GPCR, the method comprising: a) providing a mutant GPCR of a parent GPCR, wherein the mutant GPCR has increased stability in a particular conformation relative to the parent GPCR; b) providing a test compound; c) determining whether the test compound binds to the mutant GPCR when residing in a particular conformation; and d) isolating a test compound that binds to the mutant GPCR when residing in the particular conformation. Methods of producing GPCRs with increased stability relative to a parent GPCR are also disclosed. La présente invention concerne un procédé permettant de produire un partenaire de liaison spécifique conformationnel d'un GPCR (GPCR : récepteur couplé aux protéines G), ledit procédé consistant à : a) se procurer un GPCR mutant issu d'un GPCR parent, le GPCR mutant ayant une stabilité accrue dans une conformation particulière par rapport au GPCR parent; b) se procurer un composé candidat; c) déterminer si le composé candidat se lie au GPCR mutant lorsqu'il se trouve dans une conformation particulière; et d) isoler un composé candidat qui se lie au GPCR mutant lorsqu'il se trouve dans cette conformation particulière. L'invention concerne également des procédés permettant de produire des GPCR qui ont une stabilité accrue par rapport à un GPCR parent.
