RICHARDSON-MERRELL Inc 17 Dec 1973 [21 Dec 1972] 58261/73 Heading C2C Novel compounds of Formulµ I and II wherein n is an integer of from 2 to 6; R and R 1 are each hydrogen, lower alkyl having from 1 to 6 carbon atoms, cycloalkyl having from 3 to 6 carbon atoms, alkenyl having from 3 to 6 carbon atoms in which the unsaturation is in a position other than in the 1-position of the alkenyl group, and when R and R 1 are taken together with the nitrogen atom to which they are attached represent the pyrrolidinyl, piperidino or morpholino radical; Z is oxygen, nitrogen and methylene; Y is nitrogen or methylidene; R 2 is hydrogen or lower alkyl having from 1 to 4 carbon atoms; and the pharmaceutically acceptable acid addition salts thereof, are prepared by one of the following methods: (a) where Z is methylene by action of diazomethane on a fluorene of Formula III: and where Y is methylidene by further action of a diazoalkane; (b) where Z is oxygen by esterification of the compound of Formula VIII: with halogen(CH 2 ) n NRR 1 ; (c) where Z is NH by reaction of sodium azide on a fluorene of Formula III above and where Y is nitrogen by introduction of the ether group R 2 . Fluorenes of Formula III above are prepared by reaction of HO-(CH 2 ) n NRR 1 with 9-oxofluorene-2,7-dicarboxylic acid chloride. The dicarboxylic acid VIII above is prepared by action of H 2 O 2 in concentrated H 2 SO 4 on 9-oxo-fluorene-2,7-dicarboxylic acid. Anti-viral compositions in conventional forms for oral, parenteral or topical administration comprise an above novel compound and excipients.
