Zusammenfassung: | Identifying and quantifying the micro-organisms useful in processes of biomining industry, comprises: extracting the DNA from sample; quantifying the extracted DNA; optionally performing a primary polymerization chain reaction (PCR) with universal starters for the domains Bacteria and/or Archaea, to amplify a genome region; performing quantitative PCR (qPCR), while employing the extracted DNA or the amplified product and the specific starters for each taxon to be determined, as sample matrix; and calculating the number of micro-organisms in each of analyzed taxons. Method to identify and quantify the micro-organisms useful in processes of biomining industry, comprises: extracting the DNA from the sample; quantifying the extracted DNA; optionally performing a primary polymerization chain reaction (PCR) with universal starters for the domains Bacteria and/or Archaea, to amplify a genome region; performing quantitative PCR (qPCR), while employing the extracted DNA or the amplified product obtained in the primary PCR and the specific starters for each taxon to be determined, as sample matrix, where the taxons are Total bacteria, Acidiphilium sp., Leptospirillum sp., Sulfobacillus sp., Acidithiobacillus ferrooxidans and Acidithiobacillus thiooxidans and Total Archeans, Acidianus sp., Ferroplasma sp., Metallosphaera sp., Sulfolobus sp. and Thermoplasma sp.; and calculating the number of micro-organisms in each of analyzed taxons present in the sample. |
