Zusammenfassung:We studied the effect of HCMV (AD-169) on the regulation of PAR1 reporter activity in HUVEC. In HUVEC, HCMV significantly induced activation of PAR1 reporter constructs carrying AP-2-like and two complexes of AP-2/Sp1-binding elements. Shorter constructs with no AP-2-like binding elements had lower activity upon virus inoculation. Moreover, activity of the shortest construct without binding elements was not significantly increased in virus-exposed cells. Furthermore, in HCMV-exposed cells, activity of the PAR1 reporter was significantly augmented upon p52/RelB transfection. Taken together, the data indicate that activity of the PAR1 promoter is highly dependent on AP-2/Sp1 bind?ing elements and mediated by p52/RelB regulatory factors. Key words: HCMV, PAR1 gene reporter, HUVEC. Autori su u ovoj studiji analizirali uticaj HCMV (AD-169) na regulaciju aktivnosti PAR1 promotora u HUVEC. U ovim celijama, izlaganje virusu je znacajno povecalo aktivnost PAR1 reporter konstrukata koji nose AP-2-slicne vezujuce elemente zajedno sa AP-2/Sp1-vezujucim elementima organizovanim u dva kompleksa. Aktivnost progresivno kracih konstrukti koji nemaju AP-2-slicne vezujuce elemente je bila znatno smanjena posle izlaganja virusu, ali i dalje znacajno veca od njihovih osnovnih aktivnosti izmerenih u celijama koje nisu bile u dodiru sa virusom. Medjutim, aktivnost najkraceg konstrukta, koji ne poseduje niti jedan vezujuci element, nije bila znacajno povecana u celijama izlozenim virusu niti je aktivnost bila znacajno povecana u odnosu na osnovnu aktivnost samog konstrukta. Autori su jos pokazali da u celijama izlozenim virusu, transkripcioni faktori p52/RelB znacajno povecavaju aktivnost PAR1 reportera. Autori na kraju diskusije zakljucuju da je aktivnost PAR1 promotora u velikoj meri zavisna od prisustva AP-2/Sp1-vezujucih elemenata, ali i da transkripcioni faktori p52/RelB znacajno regulisu aktivnost promotora u celijama izlozenim virusu.
