Zusammenfassung:Objective To investigate the effect of siRNA mediated KDM3A gene silencing on cell proliferation and apoptosis of breast cancer cell line MDA-MB-231. Methods Constructed a KDM3A gene-specific siRNA lentiviral vector to infect breast cancer MDA-MB-231 cells, and detected the efficiency of breast cancer cells transfected with siRNA-KDM3A by the real-time fluorescent quantitative PCR. CCK-8 method was used to detect cell proliferation, Western blot was used to test cell apoptosis and Transwell method was used to detect cell invasion. Results Compared with the control group,the expression of KDM3A mRNA was down-regulated in KDM3A-sh1, KDM3A-sh2 and KDM3A-sh3 cells, and the most efficient interference RNA was KDM3A-sh2, with the expression of mRNA decreased by 80.1%(P<0.01).After silencing KDM3A, the proliferation of MDA-MB-231 cells decreased, the apoptosis rate increased (P<0.05), and the invasion ability decreased (P<0.01).
