The relative rate of ATP hydrolysis by EcoKI is shown when supercoiled pBRsk1 (closed circles) and linear pBRsk1 (open circles) were used as substrates. Reactions took place in high salt EcoKI buffer, pH 7.9, with 5 nM DNA and 10 nM EcoKI.
