Depicted are dot plots of isolated PBMC, labeled with FLICA and 7-AAD, in the 488 nm channel after doublet discrimination. FAM-FLICA-positive and 7-AAD-positive cells were considered pyroptotic. (B) Measurement of extracellular ASC specks. Depicted are dot plots of isolated ASC-GFP particles, labeled with ASC-B3-Alexa-647 antibodies, in the 488 nm channel and 640 nm channels (APC). The resulting gate in the Alexa-647 plot was then used for the detection of ASC specks in human serum after staining with ASC-B3-Alexa-647 antibodies.
