Zusammenfassung: | Copyright information:Taken from "Differential effects of hypoxia on etoposide-induced apoptosis according to the cancer cell lines"http://www.molecular-cancer.com/content/6/1/61Molecular Cancer 2007;6():61-61.Published online 26 Sep 2007PMCID:PMC2099441. A549, MCF-7 or HepG2 cells were incubated under normoxic or hypoxic conditions with or without etoposide (50 μM) for 1 or 16 hours. , p53 was detected in total cell extracts by western blotting, using two specific anti-p53 antibodies (a monoclonal and a polyclonal). Phosphorylation of p53 on serine15 was revealed using a specific antibody. ERK2 and a-tubulin were used to assess the total amount of proteins loaded on the gel. ND = non detected , after the incubation, cells were fixed, permeabilized and stained for p53 using a specific antibody (green). Nuclei were detected with To-Pro-3 (blue). Observation was performed using a confocal microscope with the photomultiplier constant. |
