Zusammenfassung:This database is a collection of LFP data obtained in the Lab of Prof. Dr. CA Haas (Experimental Epilepsy Research, Dept of Neurosurgery, Medical Center-University of Freiburg, Faculty of Medicine, Freiburg, Germany). Eight adult transgenic male mice (C57BL/6-Tg(Thy1- eGFP)-M-Line) were injected with 50 nL of 15 mM kainate (Tocris, Bristol, UK) in the right dorsal hippocampus. Following KA injection the occurrence of a behavioral Status Epilepticus was verified. Teflon-coated platinum-iridium wires (125 mm diameter; World Precision Instruments, Sarasota, Florida, USA) were implanted 14-16 days after kainate injections at two hippocampal positions. Mice were connected to a miniature preamplifier (MPA8i, Smart Ephys/Multi-Channel Systems, Reutlingen, Germany). Signals were amplified 1000-fold, bandpass-filtered from 1 Hz to 5 kHz, and digitized with a sampling rate of 10 kHz (Power1401 analog-to-digital converter, Spike2 software, Cambridge Electronic Design, Cambridge, UK). Data was downsampled to 500 Hz and seizure detection was performed semi-automated (see https://doi.org/10.1523/ENEURO.0299-18.2019). Recording IDs are encoding mouse number, stimulation protocol, recording number, recording channel. Example: EP148_1Hz-cycle_1_HCi1_2 (mouse: EP148, stim protocol: cycle (30 min intitial LFS, then 10 min LFS off and 10 min LFS on (4x 10 min LFS), then 70 min post LFS (toal rec length 3 h), rec number: 1, position: HCi1, channel: 2). Nomenclature of recording positions: HCi1, ipsilateral dorsal hippocampus (sclerotic); HCc, contralateral dorsal hippocampus. "Pre" recodings without simtulation were obtained directly before LFS protocol and are named "pre-cycle" (Example: EP148_pre-cycle_1_HCi1_2). For detailed information see https://doi.org/10.7554/eLife.54518. Electrical Stimulation: We stimulated mice intermittently. Initially for 30 min continuously followed by 10 min ‘off’ (no stimulation) and 10 min ‘on’ (1 Hz stimulation) phase, which was repeated four times, followed by 1-hr ‘post’ recording (Paschen et al., eLife, 2020, Figure 1B, week 7). LFS: biphasic rectangular current pulses: 400 ms phase duration, ±200 mA amplitude, anodic first; MC stimulus II software, STG1004, Smart Ephys/Multi-Channel Systems, Reutlingen, Germany. Excel file contains an overview of recordings.
