Zusammenfassung: | We investigated the pharmacokinetics of Carumonam (CRMN), administered intravenously to 5 hemodialysis patients both during and between hemodialysis treatments. During the interdialysis period the serum concentration of CRMN at 15 min, 24hr after intravenous injection of 1g of CRMN was 70.0±14.7, 42.9±6.4, and 13.4±2.1μg/ml, The β phase half-life (t1/2β) was 11.6hr, the area under the concentration time curve (AUC) was 903.6μg·hr/ml and the serum clearance of CRMN (CLs) was 18.4ml/min. But during the dialysis period the serum concentration of CRMN at 15 min after intravenous injection of 1g of CRMN was 65.5±16.0μg/ml, and it rapidly declined to 16.6±6.4μg/ml at 4hr. The t1/2β was 2.8hr, the AUC was 203.65μg·hr/ml and the CLs was 81.84ml/min. From these data we calculated that 53% of the CRMN in a patient with a body weight of 50kg is removed by a 4-hr hemodialysis. The serum concentration of CRMN at 24hr after intravenous injection of 1g of CRMN was kept up to an effective serum level in hemodialysis patients. Thus a significant proportion of the CRMN is easily removed by hemodialysis, so that the CRMN has to be replenished after hemodialysis. |
