Zusammenfassung: | Present study was undertaken to reveal the effect of nucleosides and nucleotides on DNA and RNA syntheses in regenerating liver and hepatoma using rat hepatocytes in primary culture and established ascites hepatoma cell line, AH130, in floating culture. Each group was subdivided into inosine, guanosine 5' monophosphate (GMP), cytidine, uridine, thymidine and their mixture group. Various concentration of nucleosides and nucleotides was added to the medium in cultured normal hepatocytes and AH130. In normal hepatocytes, DNA and RNA syntheses were significantly increased in inosine, GMP, uridine, thymidine, and mixture groups in appropriate concentration. Cytidine group, however, was not increased in this study. DNA and RNA syntheses of AH130 were also significantly increased in the same groups, but were increased in higher concentration compared to normal hepatocytes. Therefore, low concentration of nucleosides and nucleotides is effective on regenerating liver. However, they were unfavorable in hepatoma. |
