Zusammenfassung: | We investigated the therapeutic effect of prostaglandin E1 (PGE1) on the renal toxicity seen in lung cancer patients treated with cis-diammine-dichloroplatinum (CDDP) by measuring blood urea nitrogen (BUN), serum creatinine and creatinine clearance as factors of renal toxicity. The chemotherapy regimen was CDDP, VDS, MMC or CDDP, VP-16. Daily doses of 60μ×2 PGE1 were 10-14 days by drip infusion. Six of the 7 patients showed improvement of renal function. To confirm the therapeutic effect of PGE1, an experimental pathological study was performed. Serum BUN and Cr levels after CDDP administration in ICR strain mice significantly increased. Morphologic changes were seen in proximal and distal tubules. However treatment with PGE1 reduced CDDP-induced proximal and distal tubular damage and resulted in noincrease in serum BUN and Cr. In summary, it was demonstrated that PGE1 had therapeutic effect on renal toxicity following CDDP administration, and the possibility was suggested that increased doses of CDDP could be used with combined administration of PGE1. |
